Pesterquest Cracked
I hate to burst your bubble here but this is on Beforus, so we're rewriting a good chunk of characters along people's fancanons as is because these kids are 13 years old now. Damara, Mituna, Horuss, Kurloz, Cronus; they were all very different before Openbound. We've got nothing BUT fancanons to work with over here. We're up to our nooks in fancanons.

Our job here is to flesh out the alpha trolls with a more in-depth exploration, create a game that's fun, and let longtime (or short-time! hello!) fans befriend them. In that exploration, some will possibly happen to be cis, and some will possibly not.
We appreciate how Pesterquest's theme that 'the story belongs to you' recognizes and seeks to further the way that Homestuck has been handed over to a broadening group of writers, and has been shaped so immensely by its fans.
So we, like Pesterquest, have the opportunity to make something that does better by its fans in representation—in this and other areas.
Trans and nonbinary people have, to our all-you-can-eat buffet of cis media representation, a relative handful of granola. If they can go 'oh well- I'll still relate and feel self-love through this character' a thousand times about characters implied or confirmed to be cis, we can do the same for characters later confirmed (or, in our case, represented in a fanwork) to be trans.
To answer your question: we don't know yet, because not a single route's script has been completed.
Pesterquest Cracked
If you open our game to find characters confirmed to be trans and/or nonbinary, that would not be because we threw darts at a dartboard, cracked a whip at our team, and said WE NEED TO WIN THE TRANS & NONBINARY VOTE HERE. We're not just sitting in a room and rubbing our hands, cackling to ourselves as we drag another fictional character from their place in canon to the Forever Character-Altering Pit Of Gender™️ to piss all cis people off. That's not how actually being trans works, either.
If you open our game to find characters confirmed to be trans and/or nonbinary, that would probably be some of the magnificent work and input of our team, and the experiences they bring to the table, representations of which we wouldn't be able to create nearly as well without them.
As two people who're cis ourselves, we're sure as hell not anywhere close to being the best people to talk about trans and nonbinary representation in media. But it doesn't take being underrepresented in media to understand that sometimes it's just really freaking nice to see someone reflect your identity for a change. Like how some cis fans out there want all twelve alpha trolls to reflect theirs. ;v
